Therefore, therapeutic antibodies have become a major class of new drugs developed for cancer therapy in recent years. Nowadays, the superior antibody engineering techniques used to develop therapeutic antibody drugs mainly include the mouse hybridoma technique, phage display, transgenic mouse, and single B cell technique.
Mouse Hybridoma Technique
This method involves fusing antibody-producing B cells from immunized animals with immortalized myeloma cells to create hybridoma cell lines to produce monoclonal antibodies (mAbs) with high specificity for the target antigen.
Phage Display
A library of antibody fragments is displayed on the surface of phages. The phage library is then screened against the target antigen to identify phage clones expressing antigen-specific antibodies.
Transgenic Mouse
Transgenic or humanized mice can be engineered to express human antibody genes, allowing the generation of fully human antibodies.
Single B Cell Sorting
This method involves isolating individual B cells from immunized animals or human donors and screening them for antigen-specific antibodies.
