Introduction

Autophagy is a classical mechanism of intracellular energy metabolism and self-renewal, which plays an important role in biological development and maintenance of body homeostasis. On the one hand, regulating autophagy activity can disrupt the homeostasis of tumor cell internal environment and affect the survival and metastasis of tumor cells; on the other hand, tumor cells can obtain energy through autophagy, so autophagy can help maintain the balance of tumor cell product synthesis and metabolism. Based on the above characteristics, autophagy has a dual regulatory role in tumor development, and anti-tumor drugs based on the theory of regulating autophagy activity have been used in clinical treatment. However, the molecular mechanisms of autophagy related to tumors need to be further investigated, and the autophagy regulators related to tumor therapy are still limited.

Meanwhile, anti-tumor drugs based on the theory of regulating autophagy activity have been applied in clinical treatment, and tumor inhibition by improving autophagy activity has become a new idea in tumor treatment. Alfa Cytology has established an innovative tumor microenvironment center technology platform and is developing several technologies that aims to help our global partners to explore how to regulate autophagic activity and how to develop anti-tumor drugs by regulating autophagic activity.

Key Technologies

Transmission electron microscopy. Autophagosomes belong to subcellular structures. It is a very direct method to observe the morphological changes of different stages of autophagy under transmission electron microscope. morphological features of autophagolysosomes).

Fluorescence microscopy. LC3 (microtubule-associated proteins light chain 3, MAP1LC3) runs through the entire autophagy process and is a currently recognized autophagy marker. Endogenous LC3 or GFP-LC3 can be observed by fluorescence microscope, or the detection of autophagy can be realized by using GFP-LC3 single fluorescence and mCherry-GFP-LC3 dual fluorescence indicator system.

Western Blot. When autophagy is formed, cytoplasmic LC3-I will enzymatically decompose a small segment of polypeptide, and then combine with PE to convert into membrane-type LC3-II. Therefore, Western Blot detection was used, and then the level of autophagy was estimated by the ratio of LC3-II/I. p62 is a widely studied autophagy substrate, and its expression is negatively correlated with autophagy activity. Therefore, using western blot to detect the expression of p62 protein can also evaluate the level of autophagy.
