However, the number of NSCs is very small, and it is extremely difficult to obtain NSCs, there are ethical restrictions on obtaining NSCs from embryonic stem cell induction and differentiation. Fortunately, with the development of reprogramming technology, Alfa Cytology has achieved the direct reprogramming of adult cells into NSCs or the differentiation of induced pluripotent stem cells (iPSCs) to obtain a large number of NSCs, which has enabled the transplantation of NSCs into clinical applications.
Complex brain functions cannot be realized without efficient and specific information exchange and integration between neurons, and neurotransmitters, as important small biological molecules are involved in the chemical synapse-mediated information transfer process between neurons. Traditional direct measurements do not allow for the detection of transmitters at the level of individual cells and have insufficient specificity and poor temporal and spatial accuracy.
The novel probe GRAB (GPCR activation-based sensor) can accurately detect neurotransmitter dynamics in real time with high sensitivity, specificity, and resolution. We can use it to resolve complex functions of the nervous system at high temporal and spatial scales.
We form fusion proteins by inserting circularly rearranging fluorescent proteins into the region where the G protein-coupled receptor (GPCR) binds to the ligand (neurotransmitter) with the most significant conformational change. Once a specific neurotransmitter activates the GPCR to change its conformation, the conformation of the fluorescent protein linked to it also changes, and the fluorescence intensity changes, thus indicating dynamic changes in neurotransmitter concentration.
