Stable expression cell lines refer to cell lines constructed based on a cell line that consistently overexpresses or interferes with a specific gene. The basic principle is that exogenous DNA is cloned into a vector with a certain resistance, the vector is transfected into the host cell and integrated into the host chromosome, and the vector is screened with the resistance marker contained in the vector.
Lentiviral infection-drug screening is a widely used method for constructing stable cell lines with efficient integration and a wide range of target cells. Alfa Cytology screens target cells for resistance markers on a transient transfection basis. Alternatively, high transfection efficiency viruses are used to select the appropriate drugs for cell passaging according to the resistance markers contained in the different gene vectors. This results in cell lines that can stably express the target gene in a short period.
