With the widespread application of molecular biology techniques in tumor immunology research, the study of tumor immune response, immune tolerance and immune regulation mechanism has gradually risen to the molecular level, and the study of antigen receptors and other cell membrane molecule-mediated signal transduction processes has become the main frontier of contemporary immunology research. At present, a variety of signal transduction pathways have been found to be related to the immune regulation of tumors, and the most studied ones are TLRs signaling pathway, JAK-STAT signaling pathway, T-cell activation pathway, B-cell activation pathway and so on. However, the research on immune signaling pathways in tumor microenvironment (TME) is still limited, and the specific signal transduction mechanism of single pathway is not fully clarified, and the interconnection between the pathways is even less studied. Therefore, many immune mechanisms of tumors are still hypothesized and need further research to be confirmed.
Since tumor immune response, immune tolerance and immune regulation are involved in tumorigenesis, development and regression, it is imperative to investigate the pathways and molecular mechanisms of tumor immune signaling and to find potential therapeutic targets for tumor immunotherapy. With the increasing emphasis on precision tumor therapy, more and more targeted drugs, including angiogenesis inhibitors, immunomodulators and signal transduction inhibitors, are being used in clinical practice.
