Introduction

Tumor growth depends not only on genetic alterations of malignant cells, but also on alterations of the tumor microenvironment (TME, tumor microenvironment) such as stroma, blood vessels, and infiltrating inflammatory cells. The tumor microenvironment is infiltrated by a large number of inflammatory cells, including mast cells, T cells, myeloid-derived suppressor cells (MDSC), natural killer cells (NK), tumor-associated neutrophils (TAN) and tumor-associated macrophages (TAM), tumor-associated fibroblasts (TAF) and other cell subtypes, which are important components of the tumor microenvironment and play a tumor-promoting or inhibiting important role of tumor promotion or suppression.

It has been shown that inflammation is divided into acute inflammation and chronic inflammation. Acute inflammation is the body's natural defense against damaged cells, viruses, and other harmful stimuli that rapidly initiate and help the body heal itself. Most inflammatory cells in acute inflammation kill pathogens, promote tissue repair, stop tumor growth, and thus act as cancer suppressors. Once the inflammation does not subside in time, it becomes chronic inflammation, which may lead to a series of inflammatory diseases including malignant tumors. Systemic chronic inflammation (obesity, depression, etc.) and treatment-induced chronic inflammation also promote tumorigenesis and development by affecting the immune system. Chronic inflammation has been shown to be one of the culprits of tumorigenesis and development.

The link between inflammation and tumor depends on two different phenomena. On the one hand, genes involved in cancer development stimulate inflammatory processes. On the other hand, inflammation increases the risk of cancer by creating conditions for the suppression of anti-tumor immune responses and the accumulation of favorable mutations, the growth and survival of tumor cells, the formation of trophoblastic blood vessels, the ability of tumors to invade the surrounding tissue mass, and the stimulation of metastasis. Does chronic inflammation result from the migration of previously occurring acute inflammation? Or does it arise autonomously? Thus providing guidance for new targets and strategies for tumor drug resistance.

Animal models include spontaneous tumor animal models, induced tumor animal models, transplanted tumor animal models, and human tumor xenograft tumor models. The establishment of tumor animal models can study the relationship between certain tumors and inflammation and the molecular mechanism of action, can evaluate the efficacy of anti-tumor immunotherapy, can be used as a model for anti-tumor drug screening, etc. It provides a good experimental tool for the development of anti-tumor metastatic drugs and is an excellent platform for studying tumor growth and metastasis.

The combination of multi-omics to qualitify and quantify the changes of nucleic acids, proteins, and metabolites in tumor growth, and to comprehensively explore genetic information at the molecular level, provides strong evidence for the linkage and mechanism of action between inflammation and tumor growth for further development of targeted drugs and immune drugs.
