Pancreatic Cancer Cell Characterization

Pancreatic Cancer Cell Characterization

To identify early diagnosis strategies, preventive measures, and effective interventions, it is required to understand the specific cellular and molecular mechanisms of pancreatic cancer development and progression. established cancer cell lines remain powerful tools for research in pancreatic cancer (PC). Here, we offer pancreatic cancer cell analysis and evaluation services to help researchers and professionals select the cell line that best fits their research needs.

Heterogeneity of Pancreatic Cancer Cell

There are many high-frequency gene mutations in pancreatic cancer, such as KRAS(93%), TP53 (72%), CDKN2A (30%), SMAD4 (32%), etc. As a valuable resource, the established cell lines are widely utilized. So far, a series of pancreatic cancer cell lines have been established. For different research needs, cell lines should be carefully evaluated before selection and attention should be paid to cell line identification, including in vivo and in vitro growth characteristics, phenotypic characteristics (e.g., adhesion, invasion, migration, tumorigenesis, and angiogenic potential), and genotypic status of commonly altered genes.

Fibroblasts are the most abundant cell type in TME and are primarily responsible for the deposition of extracellular matrix, which drives the physiological properties of PDAC.Fig. 1 Origin and heterogeneity of pancreatic cancer-associated fibroblasts. (Halbrook, CJ.; et al., 2023)

Pancreatic Cancer Cell Phenotypic Characterization

  • Size and morphology determination
  • Proliferation and viability assays
  • Cycle and growth kinetics analysis

Pancreatic Cancer Cell Adhesion

Cell adhesion depends on the interaction of cell surface molecules with extracellular matrix components. The high metastatic potential of pancreatic cancer highlights the importance of detecting cancer cell adhesion properties.

Pancreatic Cancer Cell Migration and Invasion Assays

Studying the ability of pancreatic cancer cells to migrate is necessary to gain insight into the biological processes that mediate metastasis. Moreover, pancreatic cancer is by nature highly aggressive, thus invasion is another important phenotype of pancreatic cancer cells. It is worth noting that migration analysis monitors cell movement, while invasion analysis monitors cell movement through specific substrates.

Pancreatic Cancer Cell Angiogenic Potential

Angiogenesis is the process by which cancer cells induce endothelial cell proliferation leading to the subsequent formation of new blood vessels, also a typical feature of tumor growth and metastasis. The angiogenic potential of pancreatic cancer cell lines was evaluated by detecting the expression of angiogenic chemokines, cytokines, and enzymes.

Pancreatic Cancer Cell Tumorigenicity

Tumorigenicity demonstrates the ability or propensity of a cancer cell line to produce tumors in vivo. Tumor formation, including frequency of occurrence and growth rate, is generally measured starting after pancreatic cancer cell injection in immunocompromised mice, followed by measurement of tumor volume and mass at necropsy. These parameters mentioned above have been commonly used to estimate tumorigenicity.

Pancreatic Cancer Cell Metabolism

Pancreatic cancer is characterized by hypoxia and nutrient deficiency caused by dense interstitial fibrosis in the tumor microenvironment. Hypoxic tumor microenvironment has a great influence on various metabolic activities of cancer cells.
